The relationship between the expression of miR-498 and miR-378h in gingival crevicular fluid and Th17/Treg cell balance in patients with chronic periodontal disease
Objective:To investigate the relationship between the expression of microRNA-498(miR-498),microR-NA-378h(miR-378h)in gingival crevicular fluid(GCF)and the balance of T helper cell 17(Th17)/regulatory T cell(Treg)in patients with chronic periodontal disease.Methods:From January 2022 to February 2023,97 patients with chronic peri-odontitis admitted to our hospital were included as research subjects.They were grouped into mild group(31 cases),moderate group(42 cases)and severe group(24 cases)according to the severity of the disease,and 97 periodontally healthy subjects who were examined in our hospital were regarded as the control group.The correlation between the expression levels of miR-498 and miR-378h in gingival crevicular fluid of patients with chronic periodontitis and Th17/Treg were measured and ana-lyzed,exploring the risk factors affecting the occurrence of chronic periodontitis and evaluating the diagnostic value of miR-498 and miR-378h for chronic periodontitis.Results:There were obvious differences in smoking history,periodontal indica-tors,Th17,Treg,Th17/Treg between the study group and the control group(P<0.05).The expression levels of miR-498 and miR-378h in gingival crevicular fluid of the study group were higher than those of the control group(P<0.05),and the ex-pression levels of miR-498 and miR-378h in gingival crevicular fluid increased with the aggravation of chronic periodontal disease(P<0.05).The expression levels of miR-498 and miR-378h in gingival crevicular fluid were positively correlated with Th17/Treg(r=0.669,0.803,P<0.05).Smoking history,Th17/Treg,miR-498,and miR-378h were influencing factors for the occurrence of chronic periodontitis(P<0.05).The AUC of gingival crevicular fluid miR-498,miR-378h,and serum Th17/Treg for diagnosing chronic periodontitis alone were 0.826,0.833,and 0.854,respectively,the AUC of miR-498,miR-378h combined diagnosis was 0.932,which was higher than that of miR-498,miR-378h,and Th17/Treg alone diagnosis(P<0.05).Conclusion:The expression levels of miR-498 and miR-378h in gingival crevicular fluid are positively correlated with serum Th17/Treg levels,and are all related to the occurrence and development of periodontitis.
