MicroRNA-193a activates β-catenin pathway and regulates the progression of temporomandibular joint osteoar-thritis
Objective:To investigate the mechanism of miR-193a in regulating the progression of temporomandibular joint osteoarthritis.Methods:Rat model of temporomandipper arthritis was constructed,and the rats were divided into Con-trol,Model,miR-NC,miR-193a and miR-193a+LiCl(Wnt/β-catenin pathway activator)groups according to the transfection conditions.The pain behaviors of the rats were examined,RT-qPCR was used to detect the expression of miR-193a in tem-poromandibular joint,and Western blot was used to detect the expression of β-catenin,matrix metalloproteinase and Nav1.7.Inflammatory cytokines were detected by ELISA.The expressions of CD68 and iNOS were detected by immunohistochemistry.Results:Overexpression of miR-193a increased the swing-head threshold,IL-10 expression level,and inhibited the expression levels of β-catenin,matrix metalloproteinase,Nav1.7,TNF-α,IL-1β,IL-6,IL-23,CD68 and iNOS in rats(P<0.05).Activa-tion of β-catenin significantly reversed the effect of overexpression of miR-193a on the above indexes(P<0.05).Conclu-sion:Overexpression of miR-193a can reduce and control the progression of temporomandibular joint osteoarthritis through β-catenin.
