目的:旨在通过双向双样本孟德尔随机化研究牙周炎(periodontitis,PD)与慢性胃炎(chronic gastritis,CG)或胃溃疡(gastric ulcer,GU)之间的因果关系.方法:PD的全基因组关联研究(genome-wide association study,GWAS)数据来自Gene-Lifestyle Interactions in Dental Endpoints(GLIDE)联盟(12289 例).FinnGen联盟提供了关于CG(7605 例)和GU(4542 例)的GWAS数据.分析主要使用逆方差加权(inverse varianceweighted,IVW)方法进行.为了明确是否有水平多效性的影响,使用了 3 种不同方法:加权中位数(weighted median,WM)、MR-Egger回归和多效性残差和异常值法(MR-pleiotropy residual sum and outlier,MR-PRESSO).结果:IVW方法确定 PD对 CG(OR=0.885,95%CI:0.725~1.081,P=0.234)和GU(OR=1.034,95%CI:0.860~1.244,P=0.720)无因果影响.同样,CG和GU对PD都没有显著的因果影响.MR-Egger和加权中位数的结果与IVW方法一致.结论:在欧洲人种中,牙周炎和慢性胃炎或胃溃疡没有因果关系,反之亦然.
Periodontitis and chronic gastritis or gastric ulcers:a two-sample Mendelian randomization analysis
Objective:This study set out to look into the causal relationship between periodontitis(PD)and either chronic gastritis(CG)or gastric ulcers(GU)by a two-sample bidirectional Mendelian randomization study.Methods:Genome-wide association study(GWAS)data of PD was obtained from Gene-Lifestyle Interactions in Dental Endpoints(GLIDE)(12289 cases).The FinnGen consortium provided the GWAS data on CG(7605 cases)and GU(4542 cases).The analysis was conducted mostly using the inverse-variance weighted(IVW)approach.To identify and account for the impact of horizon-tal pleiotropy,three different methods were used:weighted median(WM),MR Egger regression,and MR pleiotropy residual sum and outlier(MR-PRESSO).Results:IVW method established that PD had no causal effect on CG(OR=0.885,95%CI:0.725~1.081,P=0.234)and GU(OR=1.034,95%CI:0.860~1.244,P=0.720).Similarly,neither CG nor GU had a sig-nificant causal influence on PD.The outcomes of the MR-Egger and weighted median were in accordance with the IVW ap-proach.Conclusion:PD and chronic gastritis or stomach ulcers are not causally related to one another,and vice versa.
