Effects of CGF combined with Bio-Oss bone powder on osteoblast apoptosis,alveolar bone morphology and BMP2/4 in rats with alveolar bone defect
Objective:To investigate the effects of concentrated growth factors(CGF)combined with Bio-Oss bone powder on osteoblast apoptosis,alveolar bone micromorphology and BMP2/4 in rats with alveolar bone defect.Methods:Fifty SPF SD male rats were randomly divided into normal(NO)group,model(MO)group,CGF(CF)group,Bio-Oss bone powder(BS)group and CGF combined Bio-Oss bone powder(CB)group,with 10 rats in each group.Alveolar bone defect modeling was performed in MO,CF,BS and CB groups.After successful modeling,CGF treatment was given to group CF,Bio-Oss bone powder treatment was given to group BS,CGF combined with Bio-Oss bone powder treatment was given to group CB,and no treatment was given to group NO and group MO.Maxillary bone tissue was taken for HE staining and Micro-CT detection,and osteoblast apoptosis was detected by TUNEL method.The expression of BMP2/4-related proteins was detected by immunoim-printing.Results:The maxillary surface of the NO group was smooth without defects,while the maxillary surface of the MO group was obviously defective,and the new bone only regenerated a little.Compared with the MO group,the maxillary defects of the CF group,BS group and CB group were significantly alleviated,and the new bone gradually covered the defective area,and the CB group had the best repair effect.Compared with NO group,the number and volume of trabeculae in MO group were decreased,and the separation degree of trabeculae was increased(P<0.05).Compared with MO group,the number and volume of trabeculae in CF,BS and CB groups were increased(P<0.05),and the separation degree of trabeculae was de-creased(P<0.05).There was no significant difference between BS group and CF group(P>0.05),and CB group had signifi-cant change compared with BS group(P<0.05).Maxillary bone tissue structure in group NO was normal,the surface was smooth,and no obvious bone resorption and new bone formation were observed.Maxillary bone tissue fiber arrangement in group MO was irregular,obvious bone resorption was observed,and a large amount of collagen fiber hyperplasia and a small a-mount of new bone formation were observed.Compared with group MO,bone resorption in group CF,BS and CB was signifi-cantly reduced,and new bone formation was significantly increased.Compared with NO group,the apoptosis rate of bone cells in MO group was increased(P<0.05).Compared with MO group,the apoptosis rate of bone cells in CF group,BS group and CB group was decreased(P<0.05),and there was no significant difference between BS group and CF group(P>0.05),and the apoptosis rate in CB group was significantly decreased than that in BS group(P<0.05).Compared with NO group,the ex-pressions of BMP2 and BMP4 in maxillary bone tissue of MO group were decreased(P<0.05),while the expressions of BMP2 and BMP4 in maxillary bone tissue of CF group,BS group and CB group were increased compared with MO group(P<0.05),and there was no significant difference between BS group and CF group(P>0.05).CB group was significantly higher than BS group(P<0.05).Conclusion:CGF combined with Bio-Oss bone powder has a significant effect on alveolar bone de-fect rats,can effectively improve osteoblast apoptosis and alveolar bone micromorphology,and promote the expression of BMP2/4.
