结肠癌免疫微环境相关lncRNA建模分析

摘要
[目的]评估结肠癌患者的预后与免疫,建立免疫相关的长链非编码RNA(lncRNA)预后模型.[方法]通过癌症基因组图谱(TCGA)检索461个结肠癌(CC)数据集及其临床信息和转录组分析数据.通过评估免疫浸润来探索TME系统的免疫评分.结肠癌的数据集根据免疫浸润分为高分组和低分组,利用差异表达建立免疫相关lncRNA的预后模型,预测CC风险,使用wgcnaa算法识别与免疫相关lncRNAa相关的基因模块以及分子通路和网络.[结果]对免疫评分和lncrna表达进行Pearson相关性分析,发现62个免疫相关lncrna(P<0.01).运用多因素Cox回归分析计算预后模型中各lncRNA的危险比和95%CI,并对有统计学差异的的lncRNA进行K-M生存分析,得出与结肠癌预后相关的 6 个 lncRNAs(SNHG16,B4 GALT1-AS1,LINC01811,LINC02163,GAS1 RR,MIR3150BHG)并建立模型,通过评估风险率(Hazard Ratio,HR)踢除分子MIR3150BHG.此外,K-M生存分析结果显示低风险组的总生存时间较高风险组长,ROC曲线显示生存率的曲线下面积为0.742,高危组患者的总生存率比低危组患者短(P<0.001).[结论]通过生物信息学分析,成功构建了 62个免疫相关lncRNA表达水平的结肠癌预后模型,预测准确性高,确定了 5个免疫相关lncRNA(SNHG16,B4 GALT1-AS1,LINC01811,LINC02163,GAS1 RR).并分析出相关基因通路主要富集于淋巴细胞间免疫,为结肠癌研究提供新的思路.
Abstract
[Objective]To establish a prognosis model based on the immune microenvironment of long non-coding RNAs(lncRNAs)for more accurate assessment of colorectal cancer(CRC)patient prognosis.[Methods]We collected 461 colorectal cancer samples and their clinical information and transcriptome data from The Cancer Genome Atlas(TCGA)database.The ESTIMATE algorithm was used to assess immune infiltration,categorizing samples into high and low immune score groups.A prognosis model involving im-mune-related lncRNAs was established based on differential expression analysis,and the model's predictive ability was verified through clinical evaluation and decision curve analysis(DCA).[Results]Pearson correla-tion analysis identified 62 lncRNAs associated with immune scores(P<0.01).Multivariate Cox regression analysis selected 6 lncRNAs related to CRC prognosis(SNHG16,B4 GALT1-AS1,LINC01811,LINC02163,GAS1 RR,MIR3150BHG).Further analysis excluded MIR3150BHG.Kaplan-Meier survival a-nalysis showed that the low-risk group had longer overall survival,with an area under the ROC curve of 0.742,indicating that the high-risk group's overall survival was significantly shorter than that of the low-risk group(P<0.001).[Conclusion]This study successfully developed a CRC prognosis model involving 62 immune-related lncRNAs,identified 5 key lncRNAs(SNHG16,B4 GALT1-AS1,LINC01811,LINC02163,GAS1 RR),and found that related gene pathways were mainly enriched in lymphocyte immune regulation,providing a new perspective for CRC research.
