Interaction between SOX1 and OCT4 in gastric cancer and prediction of postoperative recurrence and survival of gastric cancer patients
Objective To study the interaction between sex-determining region of Y chromosome-related high mobility group box 1(SOX1)and Octamer-binding transcription factor 4(OCT4)in gastric cancer and the prediction of postoperative recurrence and survival of gastric cancer patients.Methods In this prospective study,94 patients with gastric cancer diagnosed and treated in the Central Hospital of Nanyang from January 2018 to June 2019 were selected as the research subjects.The patient's medical information were collected,and the three-year survival of patients was followed-up.Immunohistochemical staining was used to analyze the differences of protein levels of SOX1 and OCT4 between gastric cancer tissues and adjacent tissues,between patients in different recurrence groups,and between patients with different prognosis.Logistic regression analysis and ROC curve were used to study the predictive efficacy of SOX1 and OCT4 protein combined on recurrence and death of gastric cancer patients.Results The protein expression of SOX1(t=24.238,P<0.001)and OCT4(t=16.843,P<0.001)in cancer tissues were significantly lower than those in adjacent tissues;meanwhile,SOX1(t=16.565,P<0.001)and OCT4(t=12.444,P<0.001)protein expressions in the recurrence group were significantly lower than those in the non-recurrence group,and SOX1(t=11.759,P<0.001)and OCT4(t=13.854,P<0.001)protein expressions in the death group were significantly lower than those of the survival group.② Through Logistic regression analysis,it was found that the proteins expression level of SOX1 and OCT4 were closely related to prognosis(including recurrence and death)of gastric cancer patients.③ According to the ROC curve analysis,the area under curve of SOX1 and OCT4 combined diagnosis for recurrence and death was higher than that of SOX1 and OCT4 alone.For patients with recurrent gastric cancer,the cut-off values of SOX1 and OCT4 were 0.44 and 0.35,respectively.For the dead patients,the cut-off values of SOX1 and OCT4 were 0.41 and 0.42,respectively.The combined diagnosis of SOX1 and OCT4 has significantly higher diagnostic sensitivity and specificity for recurrence and death than the single diagnosis.Conclusions SOX1 and OCT4 in gastric cancer tissues have significant predictive value for postoperative recurrence and survival prognosis of gastric cancer patients,which can be used as an important reference for clinical diagnosis and prognosis determination.
