Expression and clinical significance of cysteine β-synthase in adenocarcinoma of esophagogastric junction
Objective To investigate the expression status of cystathionine β-synthase in adenocarcinoma of esophagogastric junction(AEG)and analyze the relationship between the expression level of CBS and tumor microvascular density(MVD),clinical features and prognosis.Methods A total of 243 patients with AEG who underwent tumor resection in the Fourth Hospital of Hebei Medical University were enrolled.Surgically resected wax specimens were selected to make pathological chips and for immunohistochemistry.Expression levels of CBS,vascular endothelial growth factor(VEGF)and MVD were detected,respectively.The result were used to clarify the relationship between the expression level of CBS and VEGF,tumor MVD.Furthermore,the correlation between CBS and clinical characters and prognosis were discussed.Results The expression level of CBS was 66.67%(162/243)in AEG tissues,and significantly higher than 30.45%(74/234)in adjacent normal tissues with significant statistical difference(P<0.001).The expression level of CBS was related to T stage,lymph node metastasis,smoking history,VEGF and MVD(P<0.05),but not related to age,gender,degree of differentiation,TNM stage and the history of alcohol use(P>0.05).The median disease-free survival(DFS)of patients with low CBS expression was significantly longer than that of patients with high CBS expression with significant statistical difference(63 months vs.50 months,P=0.0039).The result of Cox multiple regression analysis showed that CBS expression(HR=1.827,95%CI:1.146-2.912,P=0.011),VEGF expression(HR=2.684,95%CI:1.637-4.401,P<0.001),MVD(HR=4.064,95%CI:2.419-6.829,P<0.001)and TNM staging(HR=2.354,95%CI:1.523-3.638,P<0.001)were independent factors affecting DFS in AEG patients.Conclusion High expression of CBS may be related to tumor neovascularization,which is one of the prognostic factors of AEG and has potential application value in the diagnosis and treatment of AEG.
