Expression and clinical significance of GALNT7 in lung cancer
Objective To analyze the expression of N-acetylgalactosaminyltransferase 7(GALNT7)in lung cancer tissue and its relationship with clinicopathological characteristics and prognosis. Methods From February 2016 to February 2018,the cancer tissues and paired normal adjacent tissues samples from 90 lung cancer patients who underwent surgical resection were collected at Longhua District People's Hospital in Shenzhen.Immunohistochemical method was applied to detect the expression of GALNT7.The relative expression level of GALNT7 mRNA in lung cancer tissues and adjacent normal tissues was detected using real-time fluorescence quantitative PCR(qPCR).Kaplan-Meier method was used to analyze the relationship between GALNT7 expression and the survival of patients.Cox risk proportional regression model was applied to analyze the factors influencing the prognosis of patients. Results The high expression rate of GALNT7 protein in lung cancer tissues was 55.56%(50/90),significantly higher than 11.11%(10/90)in normal adjacent tissues, and the difference was statistically significant(P<0.05).The GALNT7 mRNA level in lung cancer tissues was 1.51±0.25,significantly higher than that in normal adjacent tissues(1.02±0.18),and the difference was statistically significant(P<0.05).The expression of GALNT7 was correlated with tumor diameter, smoking,TNM stage, differentiation, and lymph node metastasis(P<0.05).Kaplan-Meier survival analysis showed that the 5-year survival rates of patients in the low expression group of GALNT7 was obviously higher than that in the high expression group(P<0.05).Multivariate Cox regression analysis showed that lymph node metastasis and the expression of GALNT7 were independent factors for the prognosis of lung cancer patients(P<0.05).Conclusion Abnormally high expression of GALNT7 is involved in lung carcinogenesis and has the potential to serve as a prognostic predictive molecular marker for lung cancer patients.
