Expression of LRP2 in breast cancer and its clinical significance
Objective To investigate the effect of LRP2 expression on lymph node metastasis and prognosis in breast cancer.Methods Real-time fluorescence quantitative PCR(qPCR)and immunohistochemical staining were used to detect the expression of LRP2 gene and protein.The relationship between LRP2 expression and clinicopathological features of breast cancer was analyzed.Kaplan-Meier method was used to draw the survival curve,and the survival difference was tested by Log-rank.Results LRP2 was screened as a differential gene in breast cancer expression from GSE42568 data set.Immunohistochemical staining showed that the LRP2 staining score in breast cancer tissues was lower than that in adjacent normal tissues(P<0.05).The LRP2 staining score in the group with lymph node metastasis(LMP)was lower than that in the group without lymph node metastasis(LMN),and the difference was statistically significant(P<0.05).The result of qPCR showed that the expression level of LRP2 gene in LMP group was significantly lower than that in LMN group,and the difference was statistically significant(P<0.05).The expression of LRP2 protein was correlated with T stage,lymph node metastasis and TNM stage(P<0.05),but not with age and differentiation degree(P>0.05).Analysis of RNA sequencing data of breast cancer patients in the TIMER 2.0 database showed that the overall survival rate of patients with low LRP2 expression was significantly lower than that of patients with high LRP2 expression(HR=0.75,P<0.05).Conclusion The low expression of LRP2 in breast cancer tissues is associated with lymph node metastasis and poor prognosis.
