Expression and clinical significance of IL9R in bladder urothelial carcinoma
Objective To investigate the expression and clinical significance of IL9R in bladder urothelial carcinoma,and to explore the relationship between IL9R expression and immune cell infiltration.Methods GEPIA website was used to analyze the overall survival(OS)and disease-free survival(DFS)of patients with different IL9R expression in the TCGA database.A total of 118 patients with bladder urothelial carcinoma treated by surgery in our hospital were collected as research objects.The expression level of IL9R gene was detected in samples of 15 patients,and the expression of IL9R protein was detected by immunohistochemical staining after paraffin embedding in tissues of 103 patients.The relationship between IL9R protein expression and clinicopathological features of patients was analyzed.Kaplan-Meier method was used to draw survival curves for analysis,and survival differences were tested by Log-rank.Cox proportional regression analysis of risk factors affecting the prognosis of patients with bladder urothelial carcinoma.The relationship between IL9R expression and immune cell infiltration in patients with bladder urothelial carcinoma was analyzed by TIMER database.Results GEPIA online analysis showed that the OS and DFS in patients with low expression of IL9R were significantly lower than those with high expression of IL9R.The expression level of IL9R gene in bladder urothelial carcinoma tissues was significantly lower than that in paracancer tissues(P<0.05).IL9R protein expression score in cancer tissues was 4.34±2.35,which was significantly lower than 5.22±2.55 in adjacent normal tissues,with statistical significance(P<0.05).The expression of IL9R was not correlated with sex,age,differentiation degree,T stage,N stage and M stage(P>0.05).Only the TNM stage was related(P<0.05).Univariate and multivariate Cox proportional regression analysis showed that IL9R expression was an independent factor affecting OS in patients with postoperative bladder urothelial carcinoma(P=0.03).The analysis of TIMER database showed that the expression level of IL9R was positively correlated with the infiltration degree of CD8+T cells and M1 macrophages.Conclusion The expression of IL9R is down-regulated in bladder urothelial carcinoma,and low expression of IL9R indicates poor prognosis,which may affect prognosis by inhibiting the infiltration of CD8+T cells and M1 macrophages.
