Expression of PTDSS1 in ovarian cancer and its clinical significance
Objective To investigate the expression of PTDSS1 gene and protein in ovarian cancer and its clinical significance.Methods The expression level of PTDSS1 in ovarian cancer tissues and normal tissues was compared using GEPIA online database,and the relationship between PTDSS1 expression and prognosis of patients with ovarian cancer was analyzed.The expression of PTDSS1 protein in paraffin-embedded tissues was detected by immunohistochemical method in 94 patients with ovarian cancer who underwent surgical treatment in gynecology department of our hospital from 2015 to 2020.To compare the relationship between PTDSS1 protein expression and clinicopathological features of ovarian cancer.Survival curve was drawn by Kaplan-meier method,and survival difference was tested by Log-rank.Cox proportional regression model was used to analyze the prognostic factors of ovarian cancer.Results The GEPIA online database analysis showed that the relative expression level of PTDSS1 in ovarian cancer was higher than that in normal tissues(P<0.05).Immunohistochemical staining analysis showed that PTDSS1 protein level in ovarian cancer tissues was significantly higher than that in adjacent tissues(P<0.05).The expression of PTDSS1 was not correlated with age or differentiation degree(P>0.05).It was related to TNM stage,T stage and N stage(P<0.05).According to the immunohistochemical score of PTDSS1,it was found that the expression of PTDSS1 in stage Ⅰ ovarian cancer was the lowest,and the expressions of Ⅱ to Ⅳ were significantly increased compared with stage Ⅰ ovarian cancer(P<0.01).GEPIA online analysis showed that the survival time of patients with low expression of PTDSS1 gene was better than that of patients with high expression of PTDSS1 gene(P<0.05).Kaplan-Meier method showed that the median survival time of patients with high PTDSS1 expression was 18 months,which was significantly lower than 36 months of patients with low PTDSS1 expression(P<0.01).Univariate and multivariate Cox proportional regression models showed that TNM stage and PTDSS1 protein expression level affected the influencing factors of OS in postoperative ovarian cancer patients(P<0.05).Conclusion PTDSS1 is highly expressed in ovarian cancer tissues and predicts poor prognosis.
