Expression and clinical significance of SCGB2A1 in cervical squamous cell carcinoma
Objective To investigate the expression and clinical significance of SCGB2A1 in cervical squamous cell carcinoma.Methods Differential gene expression was analyzed in GEO database GSE39001 dataset.The prognosis of patients with different SCGB2A expression in TIMER database was analyzed.A total of 123 patients with cervical squamous cell carcinoma underwent surgical treatment in the First Affiliated Hospital of Hunan College of Traditional Chinese Medicine from 2015 to 2017 were collected.The expression of SCGB2A1 gene was detected by real-time quantitative fluorescent PCR(qPCR)in 25 cancer tissues and adjacent tissues.In another 98 cases,SCGB2A1 protein expression was detected by paraffin embedding and immunohistochemical staining,and the relationship between protein expression and clinicopathological features was compared.Kaplan-Meier method was used to draw survival curve analysis,and survival difference was tested by Log-rank.Cox proportional regression analysis of risk factors affecting the prognosis of patients with bladder urothelial carcinoma.Results In GSE39001 data set,SCGB2A1 was differentially expressed in cervical squamous cell carcinoma,and its expression was down-regulated.The TIMER database found that patients with low expression of SCGB2A1 had worse OS than those with high expression of SCGB2A1(P<0.05).The result of qPCR showed that the expression level of SCGB2A1 gene in cancer tissues was significantly lower than that in adjacent tissues,and the difference was statistically significant(P<0.05).Immunohistochemical staining result showed that SCGB2A1 staining score in cancer tissues was significantly lower than that in adjacent tissues,with statistical significance(P<0.05).The expression of SCGB2A1 was correlated with the differentiation degree and TNM stage(P<0.05).The median OS was 2.5 years in patients with low SCGB2A1 expression,which was lower than 3.6 years in patients with high SCGB2A1 expression,and the difference was statistically significant(P<0.01).Univariate and multivariate Cox proportional regression analysis showed that low SCGB2A1 expression was an independent risk factor for OS in patients with cervical squamous cell carcinoma(P=0.013).Conclusion The expression of SCGB2A1 in cervical squamous cell carcinoma is low,and the prognosis of patients is poor.
