Objective To investigate the the level and clinical significance of circular RNA circSMAD2(hsa_circ_0047603 and hsa_circ_0108525)in the tissues of glioma patients.Methods Eighty-five cases of glioma tissues surgically removed and pathologically confirmed in our hospital were collected.Real-time quantitative PCR was used to detect the levels of hsa_circ_0047603 and hsa_circ_0108525 in the glioma tissues using 32 normal brain tissues as a reference.Stratified analysis was conducted to investigate the relationship between levels of hsa_circ_0047603 and hsa_circ_0108525 and the clinical pathological characteristics of glioma.The value of hsa_circ_0047603 and hsa_circ_0108525 levels in the prognosis of glioma was evaluated by combining follow-up data and Cox proportional hazards regression model.Results Levels of hsa_circ_0047603 and hsa_circ_0108525 in glioma tissues were higher than those in normal tissue(P<0.001).Levels of hsa_circ_0047603 and hsa_circ_0108525 were correlated with pathological types,WHO grading,peritumoral edema and depth of infiltration(P<0.05),but not with gender and age(P>0.05).Univariate analysis showed that the levels of hsa_circ_0047603 and hsa_circ_0108525 were associated with overall survival(OS)of gliomas(P<0.05).The median OS of low level individuals was 47.0 months for hsa_circ_0047603 and hsa_circ_0108525,better than that of high level individuals at 25.5 and 35.0 months.Multivariate analysis showed that up-regulated levels of hsa_circ_0047603(HR=2.149,95%CI:1.283-3.598,P=0.006)and hsa_circ_108525(HR=1.844,95%CI:1.022-3.327,P=0.028)were independent factors for poor prognosis in glioma.Conclusion Upregulation of circSMAD2 may have value in predicting clinical outcomes in patients with glioma,especially for those with high pathological grading,and has the potential to serve as a biomarker for this tumor.
