The expression and prognostic correlation of LOC676462 in colorectal cancer were analyzed based on TCGA database
Objective To investigate the expression of lncRNA LOC676462 in colorectal cancer and its correlation with clinicopathological features and prognosis.Methods The transcriptome and clinical data of colorectal cancer RNA were downloaded from the Cancer Gene Atlas(TCGA)database,and LOC676462 expression differential analysis and survival analysis were performed.The expression level of LOC676462 pancarcinoma was analyzed by TIMER 2.0.The expression of LOC676462 was detected by real-time quantitative fluorescent PCR(qPCR),and the relationship between the expression of LOC676462 and clinicopathological characteristics of colorectal cancer patients was analyzed.Receiver characteristic operating curve(ROC)was used to analyze LOC676462 expression in the diagnosis of colorectal cancer.The overall survival(OS)curve is plotted by Kaplan-meier method.Multivariate Cox proportional regression model was used to analyze the factors affecting OS in colorectal cancer.Results The bioinformatics analysis showed that LOC676462 was highly expressed in colorectal cancer tissues.The expression level of LOC676462 in 100 colorectal cancer tissues was 3.421±2.250,which was higher than 1±0.694 in adjacent tissues,and the difference was statistically significant(P<0.05).LOC676462 expression was correlated with TNM stage,invasion depth and lymph node metastasis(P<0.05).ROC curve analysis showed that the area under the curve of LOC676462 expression in the diagnosis of colorectal cancer was 0.918(95%CI:0.882-0.955).Kaplan-Meier results showed that the 5-year overall survival rate of patients with high expression of LOC676462 was 38%,while that of patients with low expression was 65.9%,the difference was statistically significant(P<0.05).Multivariate Cox proportional regression analysis showed that TNM stage,depth of invasion,degree of tumor differentiation and LOC676462 expression were independent factors affecting OS in colorectal cancer(P<0.05).Conclusion Long non-coding RNA LOC676462 can be used as an oncogene to participate in the occurrence and development of colorectal cancer,and it may be a potential biomarker of colorectal cancer,and can be used as an indicator to predict the survival and prognosis of patients with colorectal cancer.
