Effects of DEPDC1-AS1 on migration,invasion and Wnt/β-catenin signaling pathway in non-small cell lung cancer cells
Objective To explore the function of DEPDC1 antisense RNA 1(DEPDC1-AS1)in migration,invasion and Wnt/β-catenin signaling pathway in non-small cell lung cancer(NSCLC)cells.Methods Bioinformatics analysis and qPCR were conducted to detect expression of DEPDC1-AS1 in NSCLC tissues and cell lines.H1975 cells were divided into pcDNA3.1 group and pcDNA3.1-DEPDC1-AS1 group(upregulation of DEPDC1-AS1).SPC-A-1 cells were divided into siRNA-NC group and si-DEPDC1-AS1 group(downregulation of DEPDC1-AS1).CCK-8,Wound-healing and Transwell assays were conducted to assesse cell abilities of proliferation,migration and invasion.Western blot was used to detect the protein expression of β-catenin,phosphorylated glycogen synthase kinase 3 beta(p-gsk-3β)and c-Myc.Results DEPDC1-AS1 was found to be prominently high-expressed in NSCLC tissues and cells.The proliferation,migration and invasion abilities were inhibited in siRNA-DEPDC1-AS1 group of SPC-A-1 cells,while promoted in pcDNA3.1-DEPDC1-AS1 group of H1975 cells(P<0.01).Expression of β-catenin,p-gsk-3β and c-Myc were lower in siRNA-DEPDC1-AS1 group than those in siRNA-NC group,while higher in pcDNA3.1-DEPDC1-AS1 group than those in pcDNA3.1 group(P<0.01).Conclusion DEPDC1-AS1 may promote proliferation,migration and invasion of the NSCLC cells and activate Wnt/β-catenin signaling pathway,which plays a key role in NSCLC cells'progression.
