Effects of hypaconitine on migration and invasion of gastric cancer cells through regulating LMNB1
Objective To explore the effects of hypaconitine(HC)on cell proliferation,migration and invasion of gastric cancer cells through regulating lamin B1(LMNB1).Methods qPCR was used to detect the expression of LMNB1 in the normal human gastric epithelial GES-1 cells and human gastric cancer cell lines(MKN-1,AGS,SGC-7901,MGC-803 and MKN-45).MKN-45 cells were cultured and divided into shNC group(negative control),shLMNB1 group(downregulation of LMNB1),Control group(0 μmol/L HC),HC group(4 μmol/L HC),HC+pcDNA3.1-LMNB1 group(4 μmol/L HC and over-regulation of LMNB1).CCK-8,scratch and Transwell assays were applied to evaluate the cells'proliferation,migration and invasion,respectively.Expressions of matrix metallopeptidase-9(MMP-9)and matrix metallopeptidase-2(MMP-2)were measured by qPCR and Western blot.Results LMNB1 expressions were upregulated in both gastric cancer tissues and cell lines.Silence of LMNB1 inhibited the proliferation,migration and invasion abilities of MKN-45 cells.HC inhibited the mRNA and protein expression levels of LMNB1 in MKN-45 cells(P<0.01).The cells'proliferation,migration and invasion of HC group were decreased compared with Control group.While the cells'proliferation,migration and invasion of HC+pcDNA3.1-LMNB1 group were increased compared with HC group.Moreover,HC group showed declined expression levels of MMP-9 and MMP-2 as compared with Control group(P<0.01).HC+pcDNA3.1-LMNB1 group exhibited elevated expression levels of MMP-9 and MMP-2 as compared with HC group(P<0.01).Conclusion HC alleviated malignant behaviors of gastric cancer cells through repressing LMNB1 and doun-regulating MMP-9,MMP-2 expression,indicating that HC has anti-gastric cancer effects and good potential as a clinical treatment.
