Expression of TMEM42 in lung squamous cell carcinoma tissues and its clinical significance
Objective To investigate the expression of transmembrane protein 42(TMEM42)in lung squamous cell carcinoma(LSCC)tissues and its clinical significance.Methods TGGA database was used to analyze TMEM42 gene expression in LSCC tissues.Immunohistochemistry technology and real-time quantitative PCR were used to detect the expression levels of TMEM42 in 60 pairs of LSCC tissues and adjacent tissues.Relationship between TMEM42 expression and clinical pathological characteristics of LSCC was further analyzed.Kaplan-Meier Plotter platform was employed to analyze the relationship between TMEM42 expression and prognosis of LSCC patients online.Results Online analysis of TGGA database revealed that TMEM42 expression was lower in 503 LSCC tissues compared to 52 normal tissues(P<0.05).The integrated optical density of TMEM42 in LSCC tissues was lower than that in adjacent tissues(1.72±0.09 vs.3.83±0.61),and the mRNA level was lower than that in adjacent tissues(0.30±0.04 vs.1.06±0.06).The positivity rate of TMEM42 was lower in LSCC tissues than that in adjacent tissues[43.3%(26/60)vs.70.0%(42/60)],and the above differences were statistically significant(P<0.05).The expression of TMEM42 protein was not related to the age,gender,TNM stage,and differentiation degree of LSCC(P>0.05),but is related to lymph node metastasis(P<0.05).Among them,the lymph node metastasis rate of TMEM42 positive patients was lower than that of negative patients[42.3%(11/26)vs.70.6%(24/34)].TMEM42 expression was associated with overall survival(OS)and post-progression survival(PPS)in LSCC patients,with a median OS of 67 months for those with high TMEM42 expression,which was better than 48 months for those with low expression(HR=0.79,95%CI:0.63-0.99,P=0.040),and a median PPS of 11 months for those with high TMEM42 expression,which was better than 6 months for those with low expression(HR=0.51,95%CI:0.28-0.92,P=0.023).Conclusion TMEM42 is lowly expressed in LSCC tissues and is associated with lymph node metastasis and poor prognosis.This factor has the potential to serve as a diagnostic and therapeutic target for LSCC.
