Serum miR-152-5p and miR-503-5p expression levels in colon cancer patients and their relationship with prognosis
Objective To investigate the expression levels of serum microRNA-152-5p(miR-152-5p)and microRNA-503-5p(miR-503-5p)of colon cancer patients and their relationship with clinicopathological features and prognosis.Methods The clinical data of 120 colon cancer patients and 120 healthy subjects from July 2017 to July 2020 were collected.The expression levels of serum miR-152-5p and miR-503-5p were detected by real-time quantitative PCR(qPCR).The correlation between the expression levels of the two items was analyzed by Pearson's method,and the relationship between the expression levels and prognosis was analyzed by Kaplan-Meier method.The factors affecting the prognosis of colon cancer patients were analyzed by Cox proportional hazards regression model.Results The relative expression level of miR-152-5p in colon cancer patients was 1.73±0.22,higher than 0.99±0.19 of the control group(P<0.05).The relative expression level of miR-503-5p in colon cancer patients was 0.61±0.10,lower than 0.98±0.11 of the control group(P<0.05).The expression levels of serum miR-152-5p and miR-503-5p of colon cancer patients were negatively correlated(r=-0.411,P<0.05).The expression levels of miR-152-5p,miR-503-5p,TNM staging,lymph node metastasis,and differentiation degree were related to prognosis(P<0.05),while those of gender,age,tumor diameter,and adjuvant radiotherapy and chemotherapy were not(P>0.05).The 3-year survival rate of colon cancer patients with high expression of miR-152-5p was 40.6%,lower than 57.1%of the low expression group(P=0.004);the 3-year survival rate of the group with high expression of miR-503-5p was 59.6%,higher than 39.7%of the low expression group(P=0.001).Cox regression analysis showed that miR-152-5p,miR-503-5p,lymph node metastasis,differentiation degree,and TNM stage were independent prognostic factors affecting the survival of colon cancer patients(P<0.05).Conclusion The expression of miR-152-5p is upregulated and that of miR-503-5p is downregulated in colon cancer patients,and the two expression levels are negatively correlated.They may be related to the prognosis of colon cancer patients.
