Diagnostic significance of plasma hydrogen sulfide as a gas signaling molecule in breast cancer and its correlation analysis with clinicopathological features
Objective To explore the potential value of plasma hydrogen sulfide(H2S)level in the early diagnosis of breast cancer and its correlation with clinicopathological features.Methods Ninety female breast cancer patients who visited Xiaogan Hospital Affiliated to Wuhan University of Science and Technology from June 2023 to June 2024 were prospectively selected as the experimental group,and 60 patients with benign breast tumors in the same period were selected as the control group.The fluorescence probe(ZJ-233)was used to detect the level of peripheral blood H2S at admission.The differences in plasma H2S expression between the two groups were compared.The receiver operating characteristic(ROC)curve and consistency test were used to analyze and compare the evaluation indexes of different detection methods for breast cancer.The relationship between H2S and clinicopathological features such as age,tumor stage,and lymph node metastasis in breast cancer patients was further analyzed.Results The expression of H2S could be detected in the peripheral blood of all subjects.The expression level of H2S in plasma of patients with breast cancer and benign breast tumor was 9.10±1.44 and 7.38±0.51,respectively.The H2S level in the breast cancer group was higher than that in the benign group(P<0.05).The expression level of H2S had a certain correlation with age,menopausal status,TNM stage,axillary lymph node metastasis,vascular invasion,tumor size,molecular subtype,and HER-2 status in breast cancer patients,and the difference was statistically significant(P<0.05),while it was not related to the expression of Ki-67,ER,and PR status in breast cancer patients(P>0.05).The ROC curve analysis showed that the area under the curve(AUC)of H2S was 0.936,with a sensitivity of 80%and a specificity of 90%.The Kappa consistency between H2S detection and histopathological results was 0.677.Conclusion Compared with patients with benign breast diseases,the plasma H2S level in breast cancer patients is significantly increased and can be used as a non-invasive biomarker for breast cancer diagnosis.Breast cancer patients with larger tumor diameters,later TNM stages,axillary lymph node metastasis,and vascular invasion express higher levels of H2S,and the plasma H2Slevel in HER2-positive breast cancer patients is significantly higher than that in other molecular subtypes.Detecting the plasma H2S level can provide a reference for subsequent diagnosis,treatment,and prognostic evaluation of patients.
