Transcriptomic study on the potential biological mechanism of peptide deacetylase in cervical cancer
Objective To investigate the expression of peptide deacetylase(PDF)in cervical cancer and its correlation with prognosis through the TCGA database,and to explore its potential biological mechanisms in cervical cancer.Methods The transcriptome data and clinical data of cervical cancer samples from the TCGA database was downloaded.PDF difference analysis,survival analysis,independent prognosis analysis and correlation analysis with clinicopathological parameters were performed.The expression level of pan cancer in PDF was analyzed by TIMER 2.0.Co-expression analysis and GSEA enrichment analysis were used to identify the genes,pathways and cell functions associated with PDF in cervical cancer.Fresh cancer tissues and adjacent tissues of 50 patients with cervical cancer who underwent surgical treatment in Shaanxi Provincial People's Hospital from January 2023 to October 2023 were collected and tissue RNA was extracted.Real-time fluorescence quantitative PCR(qPCR)was used to detect the expression of PDF and RAB5IF in tissues.Results TCGA database showed high expression of PDF in cervical cancer.Survival analysis showed that the survival time of patients with low PDF expression was significantly longer than that of those with high PDF expression(P<0.001).The results of the subject working characteristic curve showed that the area under the curve of PDF expression predicting the 1,3 and 5 year overall survival(OS)rate of cervical cancer patients was 0.681,0.668 and 0.698,respectively.The expression of PDF was correlated with the grade and invasion degree of cervical cancer(P<0.05).Univariate and multivariate Cox proportional regression analysis showed that PDF was an independent prognostic factor for cervical cancer OS(P<0.001),with hazard ratios of 2.555(95%CI:1.601-4.076)and 2.622(95%CI:1.551-4.433),respectively.Pearson correlation analysis showed that RAB5IF had the most significant correlation with PDF(r=0.8,P<0.001).The expression levels of PDF and RAB5I in 50 cases of cervical cancer were higher than those in adjacent tissues(P<0.05).Pearson correlation analysis showed that PDF was positively correlated with RAB5IF(r=0.64,P<0.001).Conclusion High expression of PDF in cervical cancer is associated with poor prognosis,suggesting that PDF may be a new tumor marker and target for cervical cancer.
