Expression and clinical significance of TNNT3 in patients with acute myeloid leukemia
Objective To investigate the expression of TNNT3 in acute myeloid leukemia(AML)and its influence on prognosis.Methods The expression difference of TNNT3 gene between normal population and AML patients was analyzed by using the RNA sequencing data of AML in TCGA database.Kaplan-Meier method was used to analyze the relationship between TNNT3 expression and patient prognosis in big data AML samples.Bone marrow samples and peripheral blood samples were collected from 96 AML patients and 14 normal volunteers admitted to the People's Hospital of Xinjiang Uygur Autonomous Region,and TNNT3 expression was detected by real-time fluorescence quantitative PCR(qPCR).To analyze the relationship between TNNT3 expression and clinicopathological features of AML patients.Survival curve was drawn by Kaplan-Meier method,and survival difference was tested by Log-rank.Cox proportional regression model was used to analyze the factors affecting overall survival of AML patients.Results The results of TCGA database analysis showed that the expression of TNNT3 in AML patients was significantly higher than that in normal samples(P<0.05),and patients with high expression of TNNT3 had poor prognosis(P<0.001).The expression level of TNNT3 in 96 AML patients was 8.62±4.607,higher than that of normal volunteers(1.07±0.519),and the difference was statistically significant(P<0.01).The expression of TNNT3 was correlated with TP53 mutation,and the differences were statistically significant(P<0.01).The median survival time of patients with high expression of TNNT3 was 3.3 years,and that of patients with low expression was 4.5 years,the difference was statistically significant(P<0.01).Cox proportional regression model showed that TNNT3 expression,karyotype and TP53 mutation were independent factors affecting overall survival of AML patients(P<0.001).Conclusion The expression of TNNT3 in AML is significantly high,which is related to karyotype and TP53 mutation,and the high expression of TNNT3 predicts poor prognosis of patients.
