目的 检测急性髓系白血病(AML)中TNNT3 表达及其对预后的影响.方法 利用TCGA数据库中AML的RNA测序数据,分析正常人群和AML患者样本中TNNT3 基因的表达差异.Kaplan-Meier法分析在大数据AML样本中TNNT3 表达与患者预后的关系.收集新疆维吾尔自治区人民医院收治的 96 例AML患者、14 例正常志愿者,采集骨髓样本和外周血样本,采用实时荧光定量PCR(qPCR)检测TNNT3 表达.分析TNNT3 表达与AML患者临床病理特征的关系.采用Kaplan-Meier法绘制生存曲线,生存差异行 Log-rank 检验.Cox 风险比例回归模型分析影响 AML 患者总生存的因素.结果 TCGA数据库分析结果显示,AML患者TNNT3 表达显著高于正常样本(P<0.05),且TNNT3 高表达患者预后不良(P<0.001).96 例AML患者TNNT3 表达量为 8.62±4.607,高于正常志愿者的 1.07±0.519,差异具有统计学意义(P<0.01).TNNT3 表达与TP53 突变有关,差异具有统计学意义(P<0.01).TNNT3 高表达患者的中位生存时间(OS)为3.3 年,低表达患者的中位OS为 4.5 年,差异具有统计学意义(P<0.01).Cox风险比例回归模型结果显示,TNNT3 表达、染色体核型和TP53突变是影响AML患者总生存的独立因素(P<0.001)结论 AML中TNNT3 显著高表达,其表达与染色体核型、TP53 突变有关,且其高表达预示患者预后不良.
Expression and clinical significance of TNNT3 in patients with acute myeloid leukemia
Objective To investigate the expression of TNNT3 in acute myeloid leukemia(AML)and its influence on prognosis.Methods The expression difference of TNNT3 gene between normal population and AML patients was analyzed by using the RNA sequencing data of AML in TCGA database.Kaplan-Meier method was used to analyze the relationship between TNNT3 expression and patient prognosis in big data AML samples.Bone marrow samples and peripheral blood samples were collected from 96 AML patients and 14 normal volunteers admitted to the People's Hospital of Xinjiang Uygur Autonomous Region,and TNNT3 expression was detected by real-time fluorescence quantitative PCR(qPCR).To analyze the relationship between TNNT3 expression and clinicopathological features of AML patients.Survival curve was drawn by Kaplan-Meier method,and survival difference was tested by Log-rank.Cox proportional regression model was used to analyze the factors affecting overall survival of AML patients.Results The results of TCGA database analysis showed that the expression of TNNT3 in AML patients was significantly higher than that in normal samples(P<0.05),and patients with high expression of TNNT3 had poor prognosis(P<0.001).The expression level of TNNT3 in 96 AML patients was 8.62±4.607,higher than that of normal volunteers(1.07±0.519),and the difference was statistically significant(P<0.01).The expression of TNNT3 was correlated with TP53 mutation,and the differences were statistically significant(P<0.01).The median survival time of patients with high expression of TNNT3 was 3.3 years,and that of patients with low expression was 4.5 years,the difference was statistically significant(P<0.01).Cox proportional regression model showed that TNNT3 expression,karyotype and TP53 mutation were independent factors affecting overall survival of AML patients(P<0.001).Conclusion The expression of TNNT3 in AML is significantly high,which is related to karyotype and TP53 mutation,and the high expression of TNNT3 predicts poor prognosis of patients.
