Screening and Degradation Characteristics of Lignin Degrading Strains in Ziwuling ForestArea
In order to screen high-efficient lignin-degrading strains and explore their enzymatic activity and degra-dation,alkaline lignin was used as the sole carbon source to isolate and purify decayed wood samples from Ziwuling forest area.Guaiacol coloration method and aniline blue decolorization method were used for qualitative re-screen-ing,and the screened strains were identified by morphology and ITS gene sequence analysis.The activities of lac-case(Lac),manganese Peroxidase(Mnp),Lignin peroxidase(Lip)and the degradation rate of alkaline lignin were determined.A strain named JC01 was screened and identified as Tramete shirsuta,which could produce lig-nin-degrading enzyme.The strain JC01 had the highest manganese Peroxidase activity,reaching a maximum of 120U/L on the 6th day,while it had the highest alkaline lignin degradation rate on the 7th day with a percentage of 89.2%.The morphological changes of alkaline lignin treated with JC01 for 7 days were observed by scanning elec-tron microscope.It was found that the structure of alkaline lignin after treatment with JC01 was flaky,and the sur-face was loose.The results provide high-quality strain resources for efficient degradation of lignin,and have a good application prospect in the resource utilization of agricultural wastes.
