To explore the mechanism of action of honeysuckle in treating mouse eczema through network pharmacolo-gy and animal experiments,the active components in Flos Lonicerae were obtained by using the Systems TCMSP,and their corresponding targets were searched from SwissTarget and SEA databases by using GeneCards,Malacards,OMIM and DisGeNet databases to obtain disease targets related to eczema.The intersection targets of honeysuckle and eczema proteins were obtained by Venny platform,and the enrichment analysis of Gene Ontology(GO)and KEGG signal pathway was carried out by R software.We build a component target pathway network using Cytoscape software and analyze its most effective active ingredients and a PPI network using the String platform to identify key targets.The effect of Active ingredient in honeysuckle on eczema in mice was verified by animal experiments.Thirty female BALB/c mice with a6-week SPF grade body weight of(20±2)g were selected,of which 5 were assigned to the normal control group,and the remaining 25 were used to construct an eczema model using 1%DNCB acetone olive oil.The successfully modeled mice were randomly divided into a model group,a water extract group of different con-centrations of honeysuckle,and a positive drug group using a random number table method,with 5 mice in each group.After continuous administration for one week,the changes in mouse eczema were detected using ear thickness testing,ear allergy score,and histopathological examination.According to the screening of drug efficacy,there are 6 active ingredients of honeysuckle that can penetrate the skin,and 22 intersection targets with eczema.The biological pathways for obtaining target effects of active ingredients through GO-BP analysis mainly include cell cell adhesion and regulation of white blood cells,regulation of adhesion between white blood cells and cells,adhesion of white blood cells to vascular endothelial cells,phosphorylation and modification of peptide tyrosine,positive regulation of white blood cell adhesion,positive regulation of white blood cell adhesion to vascular endothelial cells;KEGG analy-sis revealed that the main signal pathways included the formation of neutrophil extracellular trap,Rap1 signal trans-duction pathway,leukocyte migration through endothelium;PPI has obtained six core targets,including VEGFA,ICAM1,MMP9,ITGB2,EGFR,SELP and MPO.Animal experiments showed that the blank group was basically normal;The pathological changes in the model control were significant and the damage was severe;The positive drug group and high concentration group showed less damage;The damage was more severe in the medium and low con-centration groups.The difference was statistically significant(P<0.05).Honeysuckle may inhibit eczema by regula-ting the formation of neutrophil extracellular traps and signaling pathways such as Rap1.Based on network predic-tion,honeysuckle can synergistically treat eczema through multiple components,targets,and pathways,providing new ideas and theoretical references for the development of honeysuckle.
