Optimization of process conditions for preparation of amino acid water-soluble fertilizer by enzymatic hydrolysis of blood powder
In order to utilize animal blood resources,amino acid water-soluble fertilizer is prepared using blood powder as raw material.The effects of factors such as liquid material ratio,raw material pretreatment temperature and combination enzyme ratio on the efficiency of blood powder enzymatic hydrolysis are studied through single factor experiments;The effects of temperature,enzyme dosage,pH and time on the efficiency of blood powder enzymatic hydrolysis are investigated through orthogonal experiments.The results show that the optimal enzymatic hydrolysis process for blood powder are as follows:The ratio of liquid volume to material mass is 6 mUg,the temperature is 55 ℃,the addition amount of combined enzyme is 3.0%(the mass ratio of protease B to flavor protease in the combined enzyme is 2∶1),the pH is 8.0,and the time is 12 hours.Under these process conditions,the hydrolysis degree of blood powder protein is 67.26%,the mass fraction of 19 free amino acids in the enzymatic solution is 6.39%,and the proportion of peptides with molecular weight below 1046 Da reaches 99.59%.By optimizing the enzymatic hydrolysis process of blood powder,the hydrolysis degree of blood powder protein has been improved,and the amino acid content in the enzymatic hydrolysis solution has been increased,providing theoretical guidance for the process control and large-scale application of enzymatic hydrolysis of blood to prepare amino acid water-soluble fertilizer.
