目的 探讨蓝莓花色苷对氧化低密度脂蛋白(ox-LDL)诱导的血管内皮细胞损伤的影响和作用机制。方法 体外培养人脐静脉内皮细胞EA。hy926,建立ox-LDL损伤模型。将EA。hy926 细胞分为对照组(正常接种细胞但不做任何处理)、模型组(100 mg/ml ox-LDL)、低剂量组(100 mg/ml ox-LDL+100 μg/ml 蓝莓花色苷)和高剂量组(100 mg/ml ox-LDL+200 μg/ml蓝莓花色苷)。CCK8 法检测细胞增殖活性。检测细胞内丙二醛(MDA)水平和谷胱甘肽过氧化物酶(GSH-Px)活性。采用酶联免疫吸附实验检测细胞培养液中黏附因子表达水平。Western blot测定凋亡相关蛋白表达情况。结果 低剂量组、高剂量组、对照组细胞活力高于模型组,差异均有统计学意义(P<0。05)。模型组MDA水平高于对照组、低剂量组、高剂量组,差异均有统计学意义(P<0。05)。模型组GSH-Px水平低于对照组、低剂量组、高剂量组,差异均有统计学意义(P<0。05)。模型组细胞间黏附分子-1、血管细胞黏附因子-1 水平高于对照组、低剂量组、高剂量组,差异均有统计学意义(P<0。05)。模型组Bax/Bcl-2、Cleaved caspase-3/Caspase-3 表达量高于对照组、低剂量组、高剂量组,差异均有统计学意义(P<0。05)。结论 蓝莓花色苷对ox-LDL诱导的血管内皮细胞损伤具有保护作用,其机制可能与抑制细胞凋亡通路相关蛋白的表达,提高细胞内抗氧化酶活性有关。
Protective effect of blueberry anthocyanins on vascular endothelial injury induced by oxidative low density lipoprotein
Objective To investigate the effect of blueberry anthocyanins on oxidative low density lipoprotein(ox-LDL)induced vascu-lar endothelial cell injury and its mechanism.Methods EA.hy926 human umbilical vein endothelial cells were cultured in vitro.Ox-LDL injury model was established EA.hy926 cells were divided into control group(normal inoculation but no treatment),model group(100 mg/ml ox-LDL),low-dose group(100 mg/ml ox-LDL+100 μg/ml blueberry anthocyanins)and high-dose group(100 mg/ml ox-LDL+200 μg/ml blueberry anthocyanins).Cell proliferation activity was detected by CCK8 assay.The level of malondialdehyde(MDA)and the activity of glutathione peroxidase(GSH-Px)were detected.The expression level of adhesion factor in cell culture medi-um was detected by enzyme-linked immunosorbent assay.The expression of apoptosis-related proteins was determined by Western blot.Results The cell viability of low-dose group,high-dose group and control group was higher than that of model group,and the differ-ences were statistically significant(P<0.05).MDA level in model group was higher than that in control group,low-dose group and high-dose group,and the differences were statistically significant(P<0.05).The level of GSH-Px in model group was lower than that in control group,low-dose group and high-dose group,and the differences were statistically significant(P<0.05).Intercellular cell ad-hesion molecule-1 and vascular cell adhesion molecule-1 levels in model group were higher than those in control group,low-dose group and high-dose group,and the differences were statistically significant(P<0.05).The expression levels of Bax/Bcl-2 and Cleaved caspase-3/Caspase-3 in the model group were higher than those in the control group,low-dose group and high-dose group,with statisti-cal significance(P<0.05).Conclusion Blueberry anthocyanins have protective effects on OX-LDL induced vascular endothelial cell injury,and the mechanism may be related to inhibiting the expression of apoptosis-related proteins and increasing the activity of intra-cellular antioxidant enzymes.
