Protective effect of blueberry anthocyanins on vascular endothelial injury induced by oxidative low density lipoprotein
Objective To investigate the effect of blueberry anthocyanins on oxidative low density lipoprotein(ox-LDL)induced vascu-lar endothelial cell injury and its mechanism.Methods EA.hy926 human umbilical vein endothelial cells were cultured in vitro.Ox-LDL injury model was established EA.hy926 cells were divided into control group(normal inoculation but no treatment),model group(100 mg/ml ox-LDL),low-dose group(100 mg/ml ox-LDL+100 μg/ml blueberry anthocyanins)and high-dose group(100 mg/ml ox-LDL+200 μg/ml blueberry anthocyanins).Cell proliferation activity was detected by CCK8 assay.The level of malondialdehyde(MDA)and the activity of glutathione peroxidase(GSH-Px)were detected.The expression level of adhesion factor in cell culture medi-um was detected by enzyme-linked immunosorbent assay.The expression of apoptosis-related proteins was determined by Western blot.Results The cell viability of low-dose group,high-dose group and control group was higher than that of model group,and the differ-ences were statistically significant(P<0.05).MDA level in model group was higher than that in control group,low-dose group and high-dose group,and the differences were statistically significant(P<0.05).The level of GSH-Px in model group was lower than that in control group,low-dose group and high-dose group,and the differences were statistically significant(P<0.05).Intercellular cell ad-hesion molecule-1 and vascular cell adhesion molecule-1 levels in model group were higher than those in control group,low-dose group and high-dose group,and the differences were statistically significant(P<0.05).The expression levels of Bax/Bcl-2 and Cleaved caspase-3/Caspase-3 in the model group were higher than those in the control group,low-dose group and high-dose group,with statisti-cal significance(P<0.05).Conclusion Blueberry anthocyanins have protective effects on OX-LDL induced vascular endothelial cell injury,and the mechanism may be related to inhibiting the expression of apoptosis-related proteins and increasing the activity of intra-cellular antioxidant enzymes.
