Preparation and evaluation of a rat model for scalded wounds
Objective Exploring a simple method for preparing a rat scald model in the laboratory.Methods Thirty SPF grade male SD rats were randomly divided into 6 groups with 5 rats in each group.One group was the blank control group and the other 5 groups were the experimental group.A scalding device with controlled temperature,fixed pressure and area was made with colorimetric tube,thermometer and constant temperature electric water bath,so that the injury time was the only variable factor,and different depth of scalding models were established on the back of rats.The pressure and temperature of the scalded parts of the 5 groups were the same,and the scalding time was 5 seconds,10 seconds,15 seconds,20 seconds,25 seconds,respectively,to form a circular scalded surface with a diameter of 3 centimeters.The skin wound surface and the depth of the wound formation after the scalding were observed 24 hours after the scalding.Results After 10 seconds,15 seconds,20 seconds,the degree of shallow Ⅱ degrees,deep Ⅱ degrees and Ⅲ degrees of scald was divided according to the three-degree sextology method,in which the scald was shallow Ⅱ degrees after 10 sec-onds,deep Ⅱ degrees after 15 seconds,and Ⅲ degrees after 20 seconds.According to the evaluation criteria of scalding wound,the depth of scalding skin on the back of rats was positively correlated with the duration.Conclusion The scalding device made of colori-metric tube,thermometer and constant temperature electric water bath can control temperature,fixed pressure and area,and the injury time is the only variable factor.It can make shallow Ⅱ degree,deep Ⅱ degree and Ⅲ degree rat scalding model,which is convenient,economical and hygienic,easy to control the depth and area of the scalding,and has good repeatability.It is an ideal animal model of burning and scalding.
