Objective To establish a model of exercise induced muscle in-jury(EIMI)in rats,and explore the effect of exhaustive ex-ercise on skeletal muscle injury in rats through adenosine phosphate-activated protein kinase(AMPK)/silent information regulator fac-tor 2-related enzyme 1(SIRT1)/peroxisome proliferators-activated receptor γ coactivator lalpha(PGC-1 α)pathway.Methods Twelve male SD rats were randomly divided into blank control group and model group,with 6 rats in each group.In the model group,7 days of exhaustive exercise was used to establish the model.The serum levels of tumor necrosis factor α(TNF-α)and interleukin-6(IL-6)were detected by enzyme-linked immunosorbent assay(ELISA).The pathological changes of rats were observed by hematoxylin eosin(HE)and Masson staining.The expression of AMPK/SIRT1/PGC-1α in rat gastrocnemius was detected by Western blot.Results HE and Masson staining showed that the arrangement of muscle fibers in the gastrocnemius of rats in the model group was disordered,serious inflammatory infiltration occurred at the junction,nucleolar arrangement was disordered and exudated,fibrosis was obvious,and a large amount of collagen fibers were accumulated.ELISA results showed that the expression levels of TNF-α and IL-6 in serum of rats in model group were significantly higher than those in blank control group,and the difference was statistically significant(P<0.05).Western blot results showed that the expressions of AMPK,SIRT1 and PGC-1α in model group were lower than those in blank control group,with statistical significance(P<0.05).Conclusion EIMI can increase the levels of serum TNF-α and IL-6 inflammatory fac-tors,and inhibit AMPK/SIRT1/PGC-1α pathway.
