目的 建立大鼠运动性骨骼肌损伤(EIMI)模型,通过腺苷酸活化蛋白激酶(AMPK)/沉默信息调节因子2同源蛋白1(SIRT1)/过氧化物酶体增殖物激活受体γ共激活因子α(PGC-1α)通路探讨力竭运动对大鼠骨骼肌损伤的作用。方法 将12只雄性SD大鼠随机分为空白对照组与模型组,每组各6只。模型组采用大鼠7 d力竭运动方案建立模型。酶联免疫吸附测定(ELISA)法检测血清中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)水平;苏木精伊红(HE)、Masson染色观察大鼠病理学改变;Western blot检测大鼠腓肠肌组织AMPK/SIRT1/PGC-1α蛋白表达。结果 HE和Masson染色镜下发现,模型组大鼠腓肠肌组织肌纤维排列紊乱,连接处出现严重炎症浸润,核仁排列杂乱渗出,纤维化明显,胶原纤维大量堆积等。ELISA检测结果显示,模型组大鼠血清中TNF-α、IL-6的表达水平明显高于空白对照组大鼠,差异有统计学意义(P<0。05)。Western blot结果显示,模型组大鼠AMPK、SIRT1、PGC-1α蛋白表达量均低于空白对照组,差异有统计学意义(P<0。05)。结论 EIMI可增加血清TNF-α、IL-6炎性因子水平,抑制AMPK/SIRT1/PGC-1α通路。

Objective To establish a model of exercise induced muscle in-jury(EIMI)in rats,and explore the effect of exhaustive ex-ercise on skeletal muscle injury in rats through adenosine phosphate-activated protein kinase(AMPK)/silent information regulator fac-tor 2-related enzyme 1(SIRT1)/peroxisome proliferators-activated receptor γ coactivator lalpha(PGC-1 α)pathway.Methods Twelve male SD rats were randomly divided into blank control group and model group,with 6 rats in each group.In the model group,7 days of exhaustive exercise was used to establish the model.The serum levels of tumor necrosis factor α(TNF-α)and interleukin-6(IL-6)were detected by enzyme-linked immunosorbent assay(ELISA).The pathological changes of rats were observed by hematoxylin eosin(HE)and Masson staining.The expression of AMPK/SIRT1/PGC-1α in rat gastrocnemius was detected by Western blot.Results HE and Masson staining showed that the arrangement of muscle fibers in the gastrocnemius of rats in the model group was disordered,serious inflammatory infiltration occurred at the junction,nucleolar arrangement was disordered and exudated,fibrosis was obvious,and a large amount of collagen fibers were accumulated.ELISA results showed that the expression levels of TNF-α and IL-6 in serum of rats in model group were significantly higher than those in blank control group,and the difference was statistically significant(P<0.05).Western blot results showed that the expressions of AMPK,SIRT1 and PGC-1α in model group were lower than those in blank control group,with statistical significance(P<0.05).Conclusion EIMI can increase the levels of serum TNF-α and IL-6 inflammatory fac-tors,and inhibit AMPK/SIRT1/PGC-1α pathway.
