Determination of Petroleum Hydrocarbons in Marine Organisms by Ultrasonic Extraction
A method for the determination of petroleum hydrocarbons in marine organisms by pretreatment ultrasonic extraction combined with fluorescence spectrophotometry was established.After the sample was saponified with sodium hydroxide solution and anhydrous ethanol,it was extracted with dichloromethane.After the extraction liquid passed through the alumina column to remove the muscle tissue interference,nitrogen was used blown to the near dry,cyclohexane was used to fix volume,and the content of petroleum hydrocarbons in the organism was calculated by fluorescence detection.The results showed that the detection limit of petroleum hydrocarbon was 0.17 mg·kg-1,the recovery rate of spiked standard was 90.8%~95%in the content range of 20.0~25.0 mg·kg-1,and the relative standard deviation was 4.4%~4.6%.This method is more convenient to operate,has better repeatability,and can better meet the needs of the experiment.The problems that should be paid attention to in this experiment were summarized.
