Study on Extraction Process of Anthraquinone From Rhizomes of Rubia
The anthraquinones in the rhizome of Rubia were extracted by ultrasonic assisted method using ethanol as the solvent.The extraction conditions of anthraquinone were explored by single-factor tests such as sonication time,solvent dosage and solvent concentration.A Box-Behnken response surface analysis test based on single-factor experiments of three factors at three levels was applied to optimize the extraction process.The optimum conditions were as follows:ultrasonic time 31.29 min,solvent dosage 13.47 mL,solvent volume fraction 81.15%.The highest theoretical prediction extraction rate of anthraquinone in the rhizomes of Rubia was 7.64%.The verification experiments under these conditions showed that the extraction rate of anthraquinone in the rhizomes of Rubia was 7.62%,which was close to the theoretical prediction.Therefore,the optimal extraction process can be obtained by the method of response surface analysis.Finally,the structure was characterized by UV and IR spectroscopy.
