Genetic Diversity Analysis of Pholiota nameko Strains Based on ISSR and Agronomic Phenotype
Utilizing 26 strains of Pholiota nameko as experimental materials and the ISSR molecular marker technique,we conducted a genetic relationship study on the germplasm resources of Pholiota nameko at both the cellular and molecular levels using 11 ISSR-PCR reaction primers.We also carried out the correlation a-nalysis,principal component analysis,and cluster analysis on the phenotypic traits of nine types of Pholiota nameko.The results showed that the 26 strains of the test strains were classified into five groups at the similar-ity level of 0.76.The agronomic phenotypic correlation analysis showed that there were six groups of traits with significant or highly significant correlation coefficients,suggesting that most phenotypic traits influence each other.The principal component analysis showed that the first five principal components accounted for 87.366%of the cumulative contribution rate,indicating that these five principal components can reflect the basic characteristics of the nine phenotypic traits of the Pholiota nameko.From the perspective of characteristic values and contribution rates,we found that mushroom stem length,mushroom color,the number of mushroom buds,yield,and cap thickness are the main factors causing phenotypic differences in the germplasm of Pholio-ta nameko.The analysis of phenotypic traits of the germplasm resources indicated that when the genetic dis-tance was set at 10,the 26 test strain be clustered into five groups.Our findings set the stage for future devel-opment and utilization,and provide a basis for the selection and breeding of new varieties of Pholiota nameko strains.
