辣根过氧化物酶在MOF上的固定化研究

摘要
根据介孔MOF孔径与生物大分子尺寸相当以及二者存在有机相容性匹配的特点,采用介孔分布在15~50 nm的多级孔MOF材料IPD-mesoMOF-3为载体成功实现了对辣根过氧化物酶的固定化,并评价其酶活性.紫外可见光谱、氮气物理吸附及X射线粉末衍射实验结果表明:多级孔材料IPD-mesoMOF-3可以有效吸附辣根过氧化物酶,实验固载量为28.1 mg·g-1;在中性条件下,运用邻苯二胺作为色原底物,用固定化辣根过氧化酶催化其与过氧化氢反应,获得良好的酶催化效果,说明IPD-mesoMOF-3是辣根过氧化物酶较为理想的固载材料.
Abstract
Horseradish peroxidase (HRP) is successfully immobilized on hierarchical porous material,IPD-mesoMOF-3,owing to compatibility both for the size and organic moiety of the enzyme matching with those of the porous material.The immobilized enzyme has been evaluated for the enzyme activity and also characterized by UV-Vis spectra,nitrogen adsorption as well as X-ray powder diffraction.The results show that excellent enzyme-catalytic behavior for the oxidation of O-phenylenediamine with hydrogen peroxide because HRP has been efficiently loaded with the saturated amount about 28.1mg/g on IPD-mesoMOF-3 in pH =7 while the structure of HKUST-1 nano-crystals remains unchanged.
