Study on the removal of nucleic acid from astaxanthin engineered bacteria
The aim of the experiment was to study the removal process of nucleic acid from Phaffia rhodozyma.Nucleic acid scavengers,nucleases,CTAB and NaCl were selected to remove nucleic acid from Phaffia rhodozyma.The optimal process of nucleic acid removal was obtained by detecting the rate of nucleic acid removal and astaxanthin concentrations in supernatant and PCR results between supernatant and bacteria.The results showed that the optimal process of nucleic acid removal was as follows:The fermentation solution was 20%concentrated,and the suspensions were mixed with 1 mL of 233 μL NaCl(6 mol/L)and 161 μL CTAB/NaCl at 65 ℃ for 30 min.The removal rate of nucleic acid was 80%.The results showed that the CTAB/NaCl method had the best effect of removing nucleic acid from Phaffia rhodozyma.
