Detection of African swine fever virus by fluorescent quantitative PCR
The aim of the experiment was to establish a fluorescent quantitative PCR detection method for ASFV.According to the nucleotide sequence of ASFV P72 gene,a fragment of ASFV P72 gene was cloned in the plasmid vector through gene cloning technology,a pair of specific primers and probes were designed,and curves were drawn through the recombinant plasmid with gene sequence.A fluorescence quantitative PCR method for detection of ASFV was developed.The results show that the method has high sensitivity,the linear relationship of the standard curve was good,the correlation coefficient reaches 0.999 8,the repeatability was better,and the coefficient of variation was small.Moreover,no amplification curves appeared with nucleic acids of other diseases as templates,which had good specificity.The results showed that the quantitative PCR method could help the detection of ASFV.
