Optimization of extraction technology of toad venom injection by orthogonal design
The purpose of the experiment was to optimize the extraction process of toad venom injection.The concentrations of indole alkaloids(calculated by 5-hydroxytryptamine hydrochloride)was taken as the index,and the water addition multiple(A),heating temperature(B)and extraction time(C)were taken as the factors.The extraction process of toad venom injection was optimized by L9(34)orthogonal test,and the extraction process parameters were determined.The data of orthogonal test are analyzed and processed by the maximum range method.The results showed that the ratio of adding water was the main factor affecting the concentrations of main components in the toad venom injection,and the best extraction process of toad venom injection was soaking in 10 times of water,heating and boiling(100 ℃),and extracting time was 4 h.Three batches of samples were prepared according to the optimum technology,and the content of indole alkaloids in toad venom injection was determined to be 9.92 mg/L by the ultraviolet visible spectrophotometry(UV)method.The research shows that the extraction process is stable and reliable,and can be used in pilot production.
