研究旨在了解广西某肉鸽场鸽圆环病毒(PiCV)Cap和Rep基因的遗传变异情况,通过PCR技术对感染PiCV的鸽组织样品克隆Cap和Rep基因并进行测序及序列分析.结果显示,克隆的Cap和Rep基因大小分别为816 bp和954 bp.核苷酸序列同源性比对分析结果显示,克隆的Cap和Rep基因与XJ1毒株核苷酸同源性最高,分别为95.5%和97.6%.遗传进化分析结果显示,Cap和Rep基因与XJ1毒株亲缘关系较近.研究表明,广西鸽群中存在PiCV流行,其Cap和Rep基因与XJ1毒株核苷酸同源性最高,亲缘关系较近,这为广西地区PiCV的诊断及防控提供了重要参考.
Cloning and genetic evolution analysis of Cap and Rep genes of pigeon circovirus in Guangxi
The purpose of the study was to understand the genetic variation of Cap and Rep genes of PiCV in a meat pigeon farm in Guangxi.Cap and Rep genes were cloned,sequenced and analyzed from pigeon tissue samples infected with PiCV by PCR.The results showed that the cloned Cap and Rep genes were 816 bp and 954 bp,respectively.The results of nucleotide sequence homology analysis showed that the cloned Cap and Rep genes had the highest nucleotide homology with XJ1 strain,which were 95.5%and 97.6%,respectively.Genetic evolution analysis showed that Cap and Rep genes were closely related to XJ1 strain.The results showed that PiCV was prevalent in Guangxi pigeon population,and its Cap and Rep gene had the highest nucleotide homology with XJ1 strain,which provided an important reference for the diagnosis and control of PiCV in Guangxi.
