Preparation and performance evaluation of colloidal gold immunochromatographic test cassette for canine Leishmania antibody
The purpose of this experiment was to establish a rapid method for detecting canine leishmaniasis. The colloidal gold immunochromatographic test paper was prepared by signal amplification technology, using leishmaniasis recombinant antigen 63 kDa glycoprotein(gp63)and mouse immunoglobulin G(IgG)antibody. The sensitivity, specificity, repeatability, and stability of this method were verified by testing the positive samples of dog Ehrlichia, dog Lyme, dog Babesia, dog cardiofilariasis(CHW),and dog leishmaniasis(LSH).The results showed that the positive samples of Ehrlichia,Lyme,Babesia, and CHW were all negative. The highest dilution gradient of LSH positive samples was 1:480.The coincidence rate with ELISA kit was 98.08%.Compared with the results of rk28 leishmania antibody test strip, the positive detection rate was higher. The results indicates that the colloidal gold immunochromatographic paper of canine leishmaniasis antibody has high sensitivity and specificity, and is suitable for the rapid diagnosis of canine leishmaniasis.
