Bioinformatics analysis of canine glucokinase regulatory protein
The purpose of the study was to investigate the structure,function and expression of canine GCKR protein.Bioinformatics methods were used to study the physicochemical properties,hydrophilicity/hydrophobicity,homology and conserved motif,signal peptide and transmembrane region,secondary structure,domain,tertiary structure,phosphorylation modification,interacting protein and GO annotation,subcellular localization,gene structure,promoter and transcriptional regulation of canine GCKR protein.The results showed that canine GCKR protein was an unstable acidic hydrophilic protein,which was closely related to horses and had no signal peptide and transmembrane region.The secondary structure consists of α-helix(47.36%),extended chain(12.00%),β-turn angle(4.48%),and random coil(36.16%),and it contains two sugar isomerase domains.The tertiary structure is constructed with horse GCKR protein as template and has high quality.There are 28 phosphorylation sites,and the interacting proteins include glucokinase,amide hydrolase domain 2,zinc finger protein 512,etc.It is involved in glucose metabolism and is mainly located in the endoplasmic reticulum.Its gene has 19 exons and 18 introns,and the promoter is located at 1 302~1 352 bp.Transcription factors such as Ncx,POU1F1a,C/EBPdelta can bind to the promoter.Studies have shown that canine GCKR protein may be involved in the pathogenesis of canine diabetes,and can be used as a potential target for developing related drugs.
