摘要
研究旨在了解辽宁省以色列急性麻痹病毒(IAPV)的遗传进化特点.试验选取成年患病蜜蜂提取IAPV并进行PCR测序,选取China-BJ(GenBank登录号:KX421583.1)基因序列设计12对引物进行全基因克隆,将获得的基因序列与GenBank已公布的19株IAPV序列进行遗传进化分析.结果显示:试验成功分离出IAPV,并将其命名为IAPV-LN株(GenBank登录号:MZ269472.1).IAPV-LN株与China-BJ和China-BJ2(GenBank登录号:MG599488.1)同属一个分支.研究表明,丹东市某蜂场患病蜜蜂中分离得到的IAPV-LN株与China-BJ2、China-BJ的亲缘关系最近,可能来自同一个原始毒株.
Abstract
The purpose of the study was to understand the genetic and evolutionary characteristics of Israel acute paralysis virus(IAPV)in Liaoning Province.IAPV was extracted from adult diseased honeybees and sequenced by PCR.Twelve pairs of primers were designed to clone the whole gene of China-BJ(GenBank accession number:KX421583.1),and the obtained gene sequence was analyzed by genetic evolution with 19 IAPV sequences published by GenBank.The results showed that IAPV was successfully isolated and named IAPV-LN strain(GenBank accession number:MZ269472.1).IAPV-LN strain belongs to the same branch as China-BJ and China-BJ2(GenBank accession number:MG599488.1).The results showed that the IAPV-LN strain isolated from diseased bees in a bee farm in Dandong had the closest genetic relationship with China-BJ2 and China-BJ,and might come from the same original strain.
