基于线粒体ATP6基因探讨3种绵羊系统发育关系

试验旨在探讨甘肃省藏羊(Tibetan sheep)、滩羊(Tan sheep)和小尾寒羊(Small-tailed han sheep)的系统发育关系,为甘肃省当地绵羊种质资源保护及合理利用提供参考.试验将藏羊、滩羊和小尾寒羊的线粒体ATP6基因部分序列扩增后送样测序,将测序结果处理后与国内外多种绵羊线粒体ATP6基因序列片段进行比对.比对完成后用DnaSP 5.0软件进行遗传多样性分析,并利用MEGA-X软件建立系统发育树,从而探讨不同绵羊种群之间的系统发育关系.结果显示,试验选择的藏羊、滩羊、小尾寒羊种群的单倍型数量分别为6、6、5个,单倍型多样性分别为0.51579、0.92857、0.93333,核苷酸多样性分别为0.00293、0.00552、0.00520.系统发育树结果显示,3种绵羊品种与国内外其他绵羊品种存在基因流.研究表明,藏羊、滩羊和小尾寒羊与选择的21种其他品种绵羊均有不同程度的亲缘关系,在开展对此3种绵羊的保种工作时要注意避免与其他品种杂交.
Study on phylogenetic relationship of three sheep based on mitochondrial ATP6 gene
The experiment aimed to explore the phylogenetic relationships among Tibetan sheep,Tan sheep,and Small-tailed Han sheep in the Gansu Province,providing a reference for the protection and rational use of local sheep germplasm resources in Gansu Province.In the experiment,partial sequences of the mitochondrial ATP6 gene from Tibetan sheep,Tan sheep,and Small-tailed Han sheep were amplified and sequenced.The sequencing results were processed and compared with mitochondrial ATP6 gene sequences from various domestic and international sheep breeds.After the comparison,genetic diversity analysis was conducted using the DnaSP 5.0 software,and a phylogenetic tree was established using the MEGA-X software to explore the phylogenetic relationships between different sheep populations.The results showed that the number of haplotype selected for the Tibetan sheep population,Tan sheep population,and Small-tailed Han sheep population were 6,6,and 5,respectively,with haplotype diversities of 0.51579,0.92857,and 0.93333,and nucleotide diversities of 0.00293,0.00552,and 0.00520,respectively.The phylogenetic tree results indicated gene flow between the three sheep breeds and other domestic and international sheep breeds.The results showed that the Tibetan sheep,Tan sheep,and Small-tailed Han sheep had different degrees of kinship with the 21 other breeds of sheep,and attention should be paid to avoiding crossbreeding with other breeds when carrying out the breeding work of these three breeds of sheep.
