Isolation,identification and phylogenetic analysis of avian infectious laryngotracheitis virus
The purpose of this study was to provide a reference for the current epidemic investigation of infectious laryngotracheitis virus(ILTV)in chickens in China and related vaccine development.Laying hens suspected to be infected with ILTV in a chicken farm in Puyang City,Henan Province were treated with positive samples and inoculated with chicken embryo chorioallasic membrane.After blind transmission for three generations,the products were identified by PCR and analyzed by TK and gD gene sequences.The pathogenicity of the isolates was preliminarily studied by animal regression test.The results showed that the chorioallantoic membrane had acne spots and thickening after three generations of blind transmission,and the product was identified positive by PCR and named PY strain.The similarity of TK and gD nucleotide sequences of PY strain to ILTV sequences published by GenBank was 99.4%~100.0%and 99.1%~100.0%,respectively.The typical clinical symptoms of infectious laryngotracheitis were found in 49-day-old SPF chickens on the 3rd day after the challenge.The morbidity and mortality of chickens were 100%and 30%respectively.The study indicates that the isolated PY strain is an ILTV with strong pathogenicity.
