CRISPR/Cas 9是近年发明的一种基因组编辑技术,该技术应用sgRNA和Cas9核酸酶能够完成DNA特定位点的识别及编辑,为人类进行基因编辑提供了全新的平台.CRISPR/Cas 9来源于细菌对抗噬菌体和质粒的免疫系统,具有简单、高效、低廉和易于操作等许多优点,因此被称为最新的第三代基因编辑技术.从其发明到现在仅仅3年多的时间,CRISPR/Cas 9已经被应用于整个生物医学领域.可以预料,CRISPR-Cas 9系统在中医药领域也将具有巨大应用前景.中医学能否把握这一机遇,对自身的发展相当重要.文章就CRISPR-Cas系统的发现、免疫防御机理的揭示、sgRNA-Cas9基因编辑工具的发明,以及研究进展做一综述,并对其在中医药领域的应用做了展望.
Research Progress of CRISPR-Cas 9 and Its Application in Research of Chinese Medicine
CRISPR/Cas 9 is a genome editing technology invented in recent years.The sgRNA and Cas9 can be used to create a DNA break at a sequence-specific target site,which provides a new platform for human gene editing.CRISPR/Cas 9 originates from the bacterial immune system against the phage and plasmid and it is a simple,efficient,inexpensive genome editing tool.It is called the latest third generation gene editing technology.It was just over three years since it appeared and CRISPR/Cas 9 has been applied to the entire biomedical field.It can be expected that CRISPR-Cas 9 system would also have a huge application prospects in traditional Chinese medicine.It is very important for the traditional Chinese medicine to grasp this opportunity.In this paper,we reviewed the discovery of the CRISPR-Cas system and immune defense mechanism,the invention of the sgRNA-Cas9 gene editing tool as well as the research progress.We also predicted its application in traditional Chinese medicine.
