Analysis of Determination of Glucose Content and Specific Chromatogram of Tibetan Medine "Manjing" by HPLC
Objective:To establish the analytical technologies of determination of glucose content and specific chromatogram of hydrolysate of the Tibetan medine "Manjing"with the pre-column derivatization HPLC method. Methods:Hydrolyse crude drug Manjing with trifluoroacetic acid (TFA), pre-column deriving the hydrolysate by the derivatization reagent 1-phenyl-3-methyl-5-pyrazolone (PMP), we analyzed specific chromatogram and determination of glucose content of hydrolysate of the Tibetan medicine "Manjing"by utilizing HPLC. Results:The content of the glucose from the crude drug "Manjing"of 13 batches varied from 10. 32% ~ 21. 43%. The 6 common peaks of specific chromatograms had stable reproducibility. Conclusion:This method is convenient and high specific, with good repeatability and stability, available to the analytical technologies of determination of glucose content and specific chromatogram, providing a scientific basis on regulating the use of the crude drug and comprehensive control of quality of Manjing.
