Effect of Rolling Method on Expressions of TGF-β1 and IL-6 after Quadriceps Femoris Injury in Rats at Different Time Points
Objective To observe the effects of the rolling manipulation on the motor function(grip strength and balance),muscle electrophysiological electricity and the expressions of transforming growth factor-β1(TGF-β1)and interleukin-6(IL-6)in serum and injured muscle tissues in vivo in rats with quadriceps injury at different intervention time points,and to ex-plore the mechanisms related to the repair of quadriceps injury in rats by the rolling manipulation and the optimal time points for the intervention of the rolling manipulation on skeletal muscle injury.Methods Eighty Wistar rats of both sexes were randomly di-vided into five large groups:2 h,24 h,48 h,72 h and 96 h groups,with 16 rats in each group,and each group was further divided into 4 subgroups:4 rats in the blank subgroup,4 rats in the model subgroup,4 rats in the treatment subgroup and 4 rats in the Yunnan Baiyao subgroup.All rats in the blank subgroup were injected with 400 μL of saline,and all rats in the model subgroup,treatment subgroup and Yunnan Baiyao subgroup were injected with 400 µL of tiger snake toxin for modeling,and after successful modeling,the team's homemade rolling massager treatment was administered at different time points:twice daily(at 10 am and 5 pm)for 3 minutes each time for 3 days.Yunnan Baiyao subgroup received aerosol treatment,with 2 sprays administered twice a day.The model and blank subgroups were reared normally.After each subgroup had completed 3 days of treatment,motor function tests were performed on the second day,followed by muscle electrophysiological tests,and finally,the blood was taken out from the abdominal aorta and from the most severely damaged area of the muscle(marked)to detect the expressions of the inflammatory factors TGF-β1 and IL-6 in the tissues using ELISA.Results The grip strength test:the treated group in each of the five major groups had better grip strength than the model group(P<0.05),blank group>treated group>Yunnan Baiyao group>model group(P<0.05).Balance beam test:The balance of the treated group in each of the five major groups was better than that of the model group(P<0.05),model group>Yunnan Baiyao group>treatment group>blank group(P<0.05).The muscle electro-physiology:in the test of electrophysiology,the mean amplitude of the quadriceps muscle in each group was compared,blank group>treatment group>Yunnan Baiyao group>model group(P<0.05).The serum and tissue levels of TGF-β1 and IL-6 in each of the five groups were lower in the treated group than those in the model group,and the inflammatory response was signif-icantly lower(P<0.05),and the serum and tissue levels of TGF-β1 and IL-6 in the 48 h group were model group>Yunnan Baiyao group>treated group>blank group(P<0.05)at different time points of the rolling manipulation of intervention.Conclu-sion After quadriceps injury in rats,repair by rolling manipulation treatment was effective except for the 2 h group.The rolling ma-nipulation intervention starting at 48 h was more effective for balance repair,muscle electrophysiology and inflammation regulation after quadriceps injury in rats.The rolling manipulation intervention starting at 96 h was the most effective for grip strength repair after quadriceps injury.
