Determination of Five isoflavone aglycones in Shegan Zhike Capsules(射干止咳胶囊)in Rat Blood and Pharmacokinetic Study by UPLC-MS/MS
Objective A quantitative method was established by using high performance liquid chromatography-mass spectrometry(HPLC-MS)to simultaneously determine the five active ingredients of tectorigenin,rrigenin,irisflorentin,iristectorigenin A and iristectorigenin B in rat plasma,and to explore the pharmacokinetic characteristics of Shegan Zhike Capsules(射干止咳胶囊)in rats.Methods Baicalein was used as an internal standard.The SD rats were subjected to a single gavage injection of Shegan Zhike Capsules.The blood samples were collected at different time points after administration,the contents of the five components were determined by electrospray ion source and multi-reaction monitoring mode,and the pharmacokinetic parameters were calculated by DAS2.0 software.Results The linear relationship of the five components was good(R>0.998),and the accuracy,precision and stability within and among batches met the requirements of biological sample analysis,and the relative recovery rate was 96.27%~113.27%.The results of pharmacokinetic studies showed that the values of Tmax,t1/2z,MRT(0-t)and Vz/F of the five components were 11.4~18.6 min,8.4~22.8 min,50.3~235.9 min and 128.5~2 913.7 L·kg-1,respectively.Conclusion The method established in this study is accurate,simple and reproducible,and can be used for the determination of the contents of the active ingredient of Shegan Zhike Capsules in rat plasma and the study of preclinical pharmacokinetics.
