Exploring Mechanism of Anchang Yuyang Decoction(安肠愈疡汤)in Treatment of Ulcerative Colitis Based on Proteomics Techniques
Objective To explore the target and regulation mechanism of ulcerative colitis(UC).Methods Eighteen rats were randomly divided into blank group,model group and treatment group.Except for the blank group,the other two groups drank dex-tran sulfate(DSS)solution freely,and the treatment group received a concentration of 1.5 g/mL Chinese medicine and behavioral observation.At the end of the experiment,the length of the colon tissue was observed,and the histopathology score(HPS)was per-formed in each group.Data-Independent Acquisition(DI A)proteomics technique analyzed the differential expression of the co-lon mucosal protein before and after modeling and after drug intervention.Results After 7 days of modeling,the rats in the model group were extremely tired with poor activity,and a large amount of fresh blood was adhered to the anus,indicating the success of modeling.After the intervention,the mental state improved,the gross blood gradually disappeared,and the pathology score de-creased.The differential protein results showed that there were 19 differential proteins regulated by Anchang Yuyang Decoction,a-mong which ATP6V1F,PRKAB1 and PDK 1 were significantly up-regulated,while the proteins of EIF2B3,KRT 17 and MCM 4 were down-regulated.The Gene Ontology(GO)annotation results showed that the differential proteins were mainly involved in biological processes such as stress,drug metabolism,negative translation and intracellular signaling,mainly located in the intracel-lular space and cell surface,and had molecular functions such as protein kinase activity,helicase activity and binding activity.Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment results showed that UC involved several signaling pathways,a-mong which the Phagosome pathway enriched the largest number of differential proteins.Conclusion The curative effect of An-chang Yuyang Decoction in the UC treatment is precise,and the mechanism of action is related to anti-inflammation,regulation of energy metabolism,inhibition of colon mucoinflammation-carcinoma transformation and targeted regulation of macrophage po-larization,and it may participate in the brain-gut interaction.
