Determination of Fecal Coliform in Sludge Based on Comparison of Enzyme Substrate Method and Multi-tube Fermentation Method
In order to verify the feasibility of enzyme substrate method for the determination of fecal coliform in sludge,120 groups of sludge samples with 4 concentration gradients were simultaneously detected by enzyme sub-strate method and multi-tube fermentation method,and the correlation and consistency of results of the two meth-ods for the detection of fecal coliform in sludge were compared by T-test.The correlation coefficient r of the two methods was 0.400~0.855,indicating that the enzyme substrate method and the multi-tube fermentation method had a strong correlation in the determination of fecal coliform in sludge.At the significance level of 0.05,P>0.05 for high concentration(greater than 1.0×105 MPN/g)and medium high concentration[(1.0×104)~(1.0×105)MPN/g]of the two methods indicated that there was no significant difference in the detection results of high concentration and medium high concentration of the two methods.P<0.05 for medium and low concentration(100~1.0×104 MPN/g)and low concentration(less than 100 MPN/g)indicated that there were significant differences between the two methods in the detection results of medium and low concentration,but the difference was acceptable at 95%confidence in-terval.Therefore,enzyme substrate method can be used for rapid detection of fecal coliform in sludge.
