α-淀粉酶水解马铃薯中淀粉提取还原糖参数优化

采用单因素实验方法及响应面法对α-淀粉酶水解马铃薯中的淀粉的酶用量、pH 值、温度及时间等条件进行了优化。通过响应面法得出最佳组合条件为 pH=6。0,酶加量 37 mg(0。37%),酶解时间5。1 h,酶解温度 66℃。在此工艺条件下马铃薯水解程度最大达到,还原糖提取率为 5。104%与还原糖提取率预测值 5。20%误差低于 2%。酶解动力学试验结果显示,在不同的底物浓度条件下,酶解速率的变化规律符合米氏方程的特征。采用双倒数作图法计算出 Km=99。13 g/L,Vmax=11。66 g/(L·h),得米氏方程式为 V=2。92[S]/(99。13+[S]),R2=0。99752,方程极显著,可很好地模拟α-淀粉酶酶解动力学过程。研究可以为资源化废弃马铃薯提供借鉴。
Optimization of Reducing Sugar Parameters for Starch Extraction From α-amylase Hydrolyzed Potato
This study was optimized by using a single factor experiment method and a response surface method to optimize the amount of amzyla amount,pH,temperature and time of the amyloid hydrolysis potato.The optimum combination conditions were obtained by the response surface method of pH 6.0,and the enzyme was added 37 mg(0.37%).The enzymatic hydrolysis time was 5.1 h,and the enzymatic solution was 66 ° C.In this process,the degree of hydrolysis of potato is maximized.The extraction rate of reducing sugar was 5.104%and the error be-tween the extraction rate of reducing sugar and the predicted value of 5.20%is less than 2%.The results of enzy-molysis kinetics test showed that the change of enzymolysis rate was consistent with the characteristics of Mi e-quation under different substrate concentrations.The Km and Vmax were calculated by the method of double recip-rocal drawing which were 99.13 g/L and 2.92 g/(L·h)separately.The Michaelis Equation is V=2.92[S]/(99.13+[S])and R2=0.99752.The equation was very significant which could be used to simulate the enzymol-ysis kinetic process of α-amylase.The research could provide a reference for resource-based potato waste.
