摘要

建立一测多评法(QAMS)同时测定悬钩木中3种成分的含量.运用高效液相色谱法(HPLC),以安捷伦SB-Aq-C18为色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.1%磷酸水溶液(17∶83,V/V)为流动相;柱温30 ℃,检测波长360 nm,流速1.0 mL/min;进样量10 μL.以咖啡酸为内参物,计算鞣花酸、金丝桃苷的相对较正因子(f),并测定其含量.结果表明:咖啡酸、鞣花酸、金丝桃苷分别在0.18~1.85 μg(r=0.9997),0.06~0.62 μg(r=0.9998),0.03~0.30 μg(r=0.9993)范围内与峰面积线性关系良好;平均加样回收率为97.66%~98.23%,RSD为1.77%~2.25%(n=9),均小于3.0%o指出了所建立的一测多评法可用于悬钩木中3种成分含量的同时测定,且方法简单、有效、结果准确.

Abstract

Three chemical constituents in Rubus sachalinensis are simultaneously determined by using quantitative analysis of multi-components by single-marker(QAMS).An high performance liquid chromatography(HPLC)method was used.The determination was performed on SB-Aq-C18(250 mm× 4.6 mm,5 μm)with acetonitrile-0.1%phosphoric acid(17∶83,V/V)as the mobile phase.The column temperature,detection wavelength,flow rate and injection volume were 30 ℃,230 nm,1.0 mL/min,10 μL,respectively.With caffeic acid as the internal stand-ard,the relative correction factors of ellagic acid and hyperoside were calculated,and then the content determina-tion were carried out.The standard curves of caffeic acid,ellagic acid and hyperoside all showed good linearity within 0.18~1.85 μg(r=0.9997),0.062~0.62 μg(r=0.9998),0.03~0.30 μg(r=0.9993)respectively.The aver-age sample recovery range was 97.66%~98.23%.RSD range was 1.77%~2.25%(n=9)with the RSD<3.0%.The established QAMS method can be used for simultaneously determining the contents of 3 components in Tibet-an medicine Rubus sachalinensis,and the method is simple,effective and accurate.
