摘要

2021年8月6日在宁夏回族自治区固原市泾源县六盘山国家森林公园内柳树树干上采集到1株野生真菌,观察该菌的子实体形态特征,对其ITS序列进行扩增、测序,并构建系统发育树,参照相关书籍记载的外观形态,鉴定出该野生真菌隶属担子菌纲(Basidiomycetes),多孔菌目(Polyporales),多孔菌科(Polyporaceae),层孔菌属(Fomes),为木蹄层孔菌(Fomes fomentarius L.ex.Fr);制备木蹄层孔菌发酵液和菌丝体,进行抗氧化活性测定,木蹄层孔菌菌丝体和发酵液对抗超氧自由基氧化能力、抗羟自由基氧化能力均表现出一定的差异,其清除率比较为:发酵液>菌丝体;对DHHP自由基清除作用、Fe3+还原力比较为:菌丝体>发酵液,其中对DHHP自由基清除作用差异明显,总体菌丝体的抗氧化活性高于发酵液.

Abstract

On August 6,2021,a wild fungus was collected from the trunk of a willow tree in Liupanshan National Forest Park,Jingyuan County,Guyuan City,Ningxia Hui Autonomous Region.The morphological characteristics of the fruiting body of the fungus were observed.Its ITS sequence was amplified and sequenced.A phylogenetic tree was constructed.Referring to the appearance of Fomes fungarius recorded in relevant books,the wild fungus was identified as Basidiomycetes,Polyporales,Polyporaceae,Fomes,and Fomes fomentarius L ex.Fr;fermentation broth and mycelium of Fomes fomentarius is prepared,and antioxidant activity is determined.The mycelium and fermentation broth of Fomes fomentarius L ex.Fr showed certain differences in their ability to resist superoxide free radical oxidation and hydroxyl free radical oxidation,and their clearance rates were in the order of fermenta-tion broth>mycelium;The scavenging effect on DHHP free radicals and Fe3+reducing power are both higher in mycelium than in fermentation broth,with significant differences in the scavenging effect on DHHP free radicals.Overall,the antioxidant activity of mycelium is higher than that of fermentation broth.
