高效液相色谱法测定黑米花色苷矢车菊素-3-O-葡萄糖苷方法的建立

本试验建立一种高效液相色谱法测定黑米花色苷矢车菊素-3-O-葡萄糖苷含量的方法.结果表明:本方法可有效分离 目标色谱峰.矢车菊素-3-O-葡萄糖苷在0.20~50.0 μg/mL浓度范围内呈良好的线性关系(R2为0.999 894),精密度RSD为0.1%,重复性RSD为0.2%~1.3%,检出限和定量限分别为0.1 mg/kg、0.3 mg/kg.矢车菊素 3 O 葡萄糖苷低、中、高浓度回收率分别为96.5%、98.4%、100.7%.该方法线性良好,准确度和精密度高,重复性和回收率较好,适用于黑米花色苷矢车菊素-3-O-葡萄糖苷的测定.
Determination of cyanidin-3-O-β-glucoside in black rice by high performance liquid chromatography
To establish a method for the determination of the content of cyanidin-3-O-β-glucosidein black rice.The results showed that this method separated cyanidin-3-O-β-glu-coside effectively.The peak area and concentration of cyanidin-3-O-β-glucosideshowed a good linearity within the concentration of 0.20~50.0 μg/mL,R2 was 0.999 894.The precisions RSD of cyanidin-3-O-β-glucoside was 0.1%,The reproducible RSD of cyanidin-3-O-β-glucoside was within the range of 0.2%~1.3%.The LODs of cyanidin-3-O-β-glucoside was 0.1 mg/kg,while the LOQs of cyanidin-3-O-β-glucosidewas 0.3 mg/kg.The average recoveries of cyanidin-3-O-β-glucosidefrom low concentration to high concentration were 96.5%、98.4%、100.7%.This method showed good reproducible,accurate and recovery,which was suit to determine the content of cyanidin-3-O-β-glucoside in black rice.
